The invention provides an isolated inflammation-related gene, the protein molecule encoded by the gene, and the use of the gene in diagnosis and treatment of inflammatory conditions.
DNA sequences which are capable of converting L-malate into L-lactate are incorporated into suitable vectors and used to transform both prokaryotic and eukaryotic hosts. The DNA sequences and vectors are particularly useful in conferring the ability to perform malolactic fermentation on wine producing yeasts. Saccharomyces cerevisiae 2514-10C/ySWl and E. coli RRI/ySWl were deposited at the A.T.C.C. on Aug. 16, 1982, and given accession Ser. Nos. 20651 and 39176, respectively.
Hybrid DNA capable of effecting controlled amplification in a host cell of a heterologous gene that determines a specific function. The hybrid DNA has the following three different regions of DNA: (1) an "amplicon" that is inducible by an external stimulus; (2) the heterologous gene (or a site for its insertion); and (3) a selectable marker enabling isolation of the transformed host cells. In order to make a desired compound coded by the heterologous gene, a vehicle including the hybrid DNA is i...
A technique suitable for cloning a cDNA having a base sequence coding for the ACTH/LPH precursor is disclosed. The invention is exemplified by the cloning of a cDNA fragment comprising a base sequence coding for the endorphin region. The fragment, hereinafter termed the endorphin gene cDNA sequence, was obtained from cultured mouse pituitary tumor cells known to produce the ACTH/LPH precursor protein.
A Salmonella gene, encoding a cytolysin, has been identified by screening for hemolysis on blood agar. The gene (slyA) is present in every strain of Salmonella examined in Shigella, and enteroinvasive Escherichia coli (EIEC) but not in other enterobacteriaceae. It is encoded near 28.5 minutes on the chromosome. A SlyA (salmolysin) has hemolytic and cytolytic activity and has a molecular weight predicted by the DNA sequence. LD.sub.50 and infection kinetics data in mice indicate that the toxin is...
Methods and compositions are provided for gene transfer to intact mammals with expression of the exogenous genetic material in the host. Mammalian host cells which are regenerative, normally highly proliferative or subject to induced proliferation, are transformed or modified in vitro with DNA capable of replication and expression in the host cell, wherein the DNA becomes incorporated into the cell. The modified cells are found to regenerate in the host with expression of the introduced DNA. Par...
This invention is a process to produce specific proteins coded for by eukaryotic (or prokaryotic) DNA in bacteria. The invention, which uses recombinant DNA techniques, produces proteins in their natural, functional state unencumbered by extraneous peptides.
The present invention provides isolated and purified polypeptide components of the mammalian circadian clock, polynucleotides that encode those polypeptides, expression vectors containing those polynucleotides, host cells transformed with those expression vectors, a process of making the polypeptide components using those polynucleotides and vectors, and processes using those polypeptides and polynucleotides.
The specification discloses: 1. Recombinant microbial cloning vehicles comprising heterologous DNA coding for the expression of mammalian hormone (e.g., somatostatin) and other polypeptides, including plasmids suited for the transformation of bacterial hosts. The latter incorporate a regulon homologous to the host in its untransformed state, in reading phase with the structural gene for the heterologous DNA; 2. Cloning vehicles coding for the microbial expression of a protein variously comprisin...
A microprocessor controlled, general purpose gene synthesizer for programmably synthesizing selected nucleotide sequences. Depending upon a programmed chemistry, metered volumes of selected bases and reagent/solvents are sequentially metered into the bottom of a reaction cell containing solid-supported nucleotides, where desired linkages are achieved in an agitated suspension. Self-refilling syringe metering and electrically initiated, pneumatically operated valving insures economical, non-conta...
